Mechanism of Action
It mitigates inflammatory responses by inhibiting proinflammatory cytokines (IL-1β, IL-6, IL-13) and Th2 chemokines through NF-κB pathway blockade. Valencene also enhances skin barrier integrity by boosting protective proteins like filaggrin and involucrin, alongside providing antioxidant benefits.

Key findings
- 01 Topical application of valencene (50 µM) in DNCB-sensitized NC/Nga mice significantly reduced atopic dermatitis-like symptoms (erythema, edema, itching, skin-fold thickening), restored decreased filaggrin expression, and lowered serum IgE, IL-1β, IL-6, and IL-13 levels in skin/splenic tissue. In vitro, it blocked Th2 and proinflammatory chemokines via NF-κB pathway.
- 02 (+)-Valencene (50 µM) induced HO-1 expression and inhibited iNOS expression, nitric oxide production, and HMGB1 release in LPS-stimulated RAW 264.7 cells, demonstrating anti-inflammatory and antioxidant activities.
