Mechanism of Action
Tetrahydromagnolol operates through a multi-faceted mechanism, acting as a potent and selective cannabinoid CB2 receptor agonist, which is associated with calming and anti-inflammatory responses. It also functions as a GPR55 antagonist. On the skin, it effectively neutralizes free radicals, such as superoxide anion and hydroxyl radicals, to mitigate inflammatory reactions. Furthermore, it precisely intervenes in melanin production by reducing its biosynthesis and accelerating the degradation of tyrosinase protein, thereby inhibiting tyrosinase maturation.

Key findings
- 01 In vitro studies show potent and selective CB2 receptor agonism with an EC50 of 0.170 µM.
- 02 In vitro data indicates GPR55 antagonism at a KB of 13.3 µM.
- 03 In vitro research demonstrates reduced melanin biosynthesis and decreased tyrosinase protein levels through inhibition of maturation and increased degradation.

Stability
Highly soluble in oil and organic solvents like ethanol and DMSO. While sparingly soluble in aqueous buffers, dissolution in ethanol prior to dilution can improve aqueous dispersion; however, aqueous solutions are not stable for more than one day. Stock solutions in DMSO require stringent storage at -80°C for up to 6 months or -20°C for 1 month, protected from light. Its enhanced lipophilicity and chemical stability are notable improvements over its precursor, magnolol.
Safety
Safety Profile
Tetrahydromagnolol has not undergone specific safety assessments by the Cosmetic Ingredient Review (CIR) or the Scientific Committee on Consumer Safety (SCCS) for cosmetic applications. The FDA typically does not pre-approve cosmetic ingredients, except for color additives, and as such, there is no specific FDA status or approval for Tetrahydromagnolol as a cosmetic ingredient. Therefore, detailed safety data for human application in cosmetics is currently limited.
