Mechanism of Action
This ingredient operates through several precise mechanisms to enhance skin health and luminosity. It notably functions as an Endothelin Antagonist, disrupting the signaling network that triggers melanin synthesis. By directly decreasing intracellular tyrosinase activity and inhibiting the gene and protein expression of key melanin-producing enzymes (TYR, TRP-1, DCT), it significantly reduces intracellular melanin content. Furthermore, it modulates complex cellular pathways, specifically suppressing melanogenesis by inhibiting the PKA/cAMP response element-binding (CREB) and MEK/extracellular signalling-regulated kinase (ERK) pathways, which in turn downregulates microphthalmia-associated transcription factor (MITF) and its downstream effects. It also facilitates the rapid transfer of melanosomes from melanocytes to keratinocytes, accelerating the natural breakdown and metabolism of pigment. Beyond its brightening capabilities, it forms a protective film on the skin's surface, enhancing moisture retention and shielding against external aggressors.

Key findings
- 01 In an in vitro study, a concentration of 0.01% (100ppm) demonstrated effective restraint of melanogenesis specifically induced by endothelin.
- 02 For anti-melanogenesis and the reduction of skin chloasma, a usage range of 0.02% to 0.10% is specifically recommended.
- 03 Laboratory studies on MNT-1 cells showed that 0.2 mg/mL significantly decreased α-MSH induced melanin content by 5.41% and ET-1 induced melanin content by 49.05%, coupled with an inhibition of intracellular tyrosinase activity.
- 04 At 0.45 mg/mL, reductions in α-MSH induced melanin content were 34.42% and ET-1 induced melanin content were 29.59% in MNT-1 cells, with continued inhibition of intracellular tyrosinase activity.
- 05 Further increasing the concentration to 0.7 mg/mL led to a 50.73% decrease in α-MSH induced melanin and a 40.12% decrease in ET-1 induced melanin in MNT-1 cells, while also boosting the colocalization correlation coefficient of TRP-1 and pan cytokeratin by 20.6%, indicative of enhanced melanosome transfer from melanocytes to keratinocytes.
- 06 Typical concentrations in broader skincare products, such as creams, serums, and lotions, range from 1% to 5% for general hydration, skin brightening, and texture improvement, with the higher end of this spectrum utilized for more intensive hyperpigmentation treatments.
