Mechanism of Action
Sargassum Thunbergii Extract operates through a sophisticated multi-pathway mechanism to enhance skin health. Its robust antioxidant capacity is demonstrated by high DPPH radical and hydroxyl scavenging activity, effectively reducing intracellular Reactive Oxygen Species (ROS) generation and upregulating crucial antioxidant enzymes such as superoxide dismutase and glutathione peroxidase, while simultaneously lowering malondialdehyde content. For anti-wrinkle efficacy, it significantly inhibits the mRNA expression of matrix metalloproteinase-1 (MMP-1) and matrix metalloproteinase-9 (MMP-9), enzymes known to degrade collagen, in addition to suppressing collagenase and elastase activities. In terms of skin brightening, the extract mitigates melanin synthesis by decreasing the mRNA levels of tyrosinase-related protein-1 (TRP-1). Furthermore, it exhibits anti-inflammatory properties by downregulating pro-inflammatory mediators including tumor necrosis factor-α, interleukin-6, cyclooxygenase-2, and inducible nitric oxide synthase, collectively contributing to its profound anti-aging potential.

Key findings
- 01 An in vitro study demonstrated a significant inhibition of matrix metalloproteinase-1 (MMP-1) mRNA expression by 58.6% and MMP-9 mRNA expression by 78.8% when B16-F0 cell lines were treated with 2 mg/mL (0.2%) of the extract, indicating potent anti-wrinkle activity.
- 02 Another in vitro investigation showed that 125 μg/mL (0.0125%) of the extract enhanced the viability of H2O2-treated L929 fibroblast cells and reduced intracellular Reactive Oxygen Species (ROS) generation, confirming its antioxidant efficacy.

Safety Profile
The Cosmetic Ingredient Review (CIR) assessment noted insufficient specific data for Sargassum Thunbergii Extract, though it is part of a broader group of brown algae-derived ingredients where the majority (68 out of 82) were deemed safe. Notably, it was not categorized among ingredients requiring further data in a re-review. An in vitro skin irritation test utilizing EpiSkin™ models found the ethanolic extract to be non-irritating at a concentration of 100 mg/mL (10%).
