Mechanism of Action
Sargassum glaucescens extract acts as a powerful antioxidant by directly scavenging free radicals and suppressing H2O2-induced reactive oxygen species (ROS) production. It stimulates cell renewal and protects dermal cells from UVA irradiation through the upregulation of key anti-oxidative genes (SOD1, GPX1) and DNA repair regulatory genes (XRCC1, ERCC6). Furthermore, it promotes the proliferation of human primary epidermal keratinocytes and reinforces the skin barrier by inducing the expression of genes such as TGM1, KRT10, and KRT14. Its hygroscopic nature helps maintain skin smoothness and suppleness, further supported by its ability to boost Natural Moisturizing Factor (NMF) production via Filaggrin (FLG) gene induction. The extract also exhibits anti-inflammatory, hypopigmenting, anti-wrinkle, and antibacterial properties.

Key findings
- 01 At 2 mg/ml, it stimulated the wound closure of CCD-966SK fibroblasts by 2.95-fold.
- 02 Concentrations between 0.25 and 1 mg/ml promoted CCD-966SK cell regeneration following UVA irradiation.
- 03 At 1 mg/ml, it induced expressions of anti-oxidative genes (SOD1, GPX1) and DNA repair regulatory genes (XRCC1, ERCC6) in UVA-irradiated CCD-966SK cells.
- 04 A concentration of 0.03125 mg/ml induced expressions of skin barrier-related genes (TGM1, KRT10, KRT14) and the NMF-promoting FLG gene in human primary epidermal keratinocytes.
- 05 It blocked H2O2-induced intracellular reactive oxygen species (ROS) generation by 77% at 0.125 mg/ml and reduced levels by 60% at 0.25 mg/ml.
- 06 It achieved 92.8% DPPH scavenging activity at 10 mg/ml.
- 07 When incorporated into collagen scaffolds, concentrations of 1-3 mg/ml demonstrated excellent antioxidant and antibacterial activity, improved structural stability, and enhanced cell proliferation without cytotoxicity, with 3 mg/ml exhibiting the highest cellular activity.
