Mechanism of Action
Santalol exerts its beneficial actions on the skin through several mechanisms. It functions as a powerful anti-inflammatory agent by suppressing cytokine and chemokine production, activating 11b-HSD1 in keratinocytes, and inhibiting the NF-κB pathway. As an antioxidant, it actively scavenges reactive oxygen species, neutralizes free radicals, and upregulates the skin's protective antioxidant enzymes. Furthermore, santalol contributes to skin brightening by inhibiting tyrosinase, the enzyme crucial for melanin production, and supports anti-aging by inhibiting collagen-degrading enzymes (MMP-1), promoting cell turnover, and enhancing skin elasticity.

Santalol is generally considered non-irritating to human skin at concentrations up to 20% based on 48-hour occlusive patch tests in healthy volunteers. However, the EU Scientific Committee on Consumer Safety (SCCS) has categorized alpha- and beta-santalol as an 'established contact allergen in humans' due to reported positive patch test reactions (in a small percentage of patients at 0.05% to 0.5% concentrations). While the risk of skin sensitization as a fragrance ingredient is typically considered low, its established allergen status suggests a moderate sensitization potential for a susceptible population.
