Mechanism of Action
This root extract primarily functions by inhibiting elastase, an enzyme responsible for degrading elastin, thereby helping to preserve skin structure and elasticity and reduce the appearance of wrinkles. Beyond its anti-elastase properties, the extract, encompassing broader Rhus semialata components, demonstrates significant antioxidant capacity, effectively scavenging reactive oxygen species (ROS) and protecting keratinocytes from induced cellular damage. It achieves this by preserving mitochondrial membrane potential and inhibiting apoptosis. The presence of gallic acid is noted as a key contributor to these protective antioxidant effects. Additionally, the extract contributes to skin moisturizing.

Key findings
- 01 Demonstrated a 33.15% inhibition of elastase activity, suggesting efficacy in preserving skin elasticity and potentially reducing wrinkles.
- 02 Achieved half-maximal effective concentration (EC50) in a DPPH antioxidant assay, indicating potent free radical scavenging capabilities. This study used an ethanolic extract which may include root components.
- 03 Showed no significant adverse effect on keratinocyte cell viability, with only a slight decrease observed at 20 μg/mL, indicating good cellular compatibility within this range. This study used an ethanolic extract which may include root components.

Stability
The root extract exhibits reduced stability when exposed to heat, light, and variations in pH, particularly when compared to the seed extract of Rhus semialata.
Safety
Safety Profile
Generally regarded as safe for cosmetic applications, with in vitro studies on keratinocytes showing no significant cytotoxicity up to 10 μg/mL and only a minor decrease in viability at 20 μg/mL. It is considered suitable for use as a cosmetic raw material. Note that a related Rhus semialata gall extract showed toxicity at higher concentrations (100 μg/mL) in a different cell type.
