Mechanism of Action
This extract exerts its biological effects primarily through 4-nerolidylcathecol, which provides robust antioxidant defense by scavenging free radicals and helps protect the skin against damage induced by UV radiation. It may also prevent the depletion of alpha-tocopherol (Vitamin E) following UV exposure. Furthermore, the extract demonstrates anti-inflammatory properties by inhibiting key molecular pathways such as cyclooxygenase-2 (COX-2) and nuclear factor kappa B (NF-κB), and has been shown to inhibit skin matrix metalloproteinases, enzymes implicated in collagen degradation.

Key findings
- 01 In studies conducted on hairless mice, a gel formulation containing 0.1% 4-nerolidylcathecol, a primary active compound from the extract, demonstrated significant antioxidant and photoprotective properties. This formulation effectively delayed the onset and reduced the incidence of UVB-induced skin damage and photoaging.
- 02 Emulsions formulated with 0.1% 4-nerolidylchatecol also exhibited potent antioxidant activity on the skin, similarly delaying the onset and reducing the incidence of UVB-induced skin damage and photoaging.

Stability
A gel formulation containing 0.1% 4-nerolidylcathecol exhibited stability without significant degradation for 103 days when stored at 5°C and 25°C. However, degradation of the gel structure was observed at 45°C. Emulsions incorporating 0.1% 4-nerolidylchatecol can be optimized for aesthetic appearance, centrifuge stability, and skin permeation, with the latter notably enhanced by the oil phase.
Synergies
- Enhanced skin permeation in emulsions when formulated with an oil phase.
Safety
Safety Profile
Direct safety assessments by regulatory bodies such as CIR, SCCS, or FDA for Pothomorphe Umbellata Root Extract are not available. However, studies indicate the absence of mutagenicity for Pothomorphe umbellata and a related species, Pothomorphe peltata, in the Salmonella/mammalian-microsome mutagenicity assay. An oral LD50 in animal models for a water-ethanolic extract of the aerial parts was found to be higher than 2.0 g/kg, suggesting low acute oral toxicity.
