POTASSIUM METHOXYSALICYLATE.

Valuable CAS 152312-71-5 / BLEACHING

Potassium Methoxysalicylate, commonly known as 4MSK, is a precision-engineered skin brightener. It effectively targets hyperpigmentation by inhibiting melanin synthesis and accelerating the skin's natural renewal process, promoting a visibly lighter and more uniform complexion. This ingredient is designed for addressing dark spots and overall skin dullness.

Bleaching Skin lightening Inhibits tyrosinase Promotes keratinization Evens skin tone

Science

Potassium Methoxysalicylate works through a dual mechanism to reduce skin pigmentation. Primarily, it suppresses melanogenesis by inhibiting tyrosinase activity within melanocytes, thereby reducing melanin production. Concurrently, 4MSK enhances epidermal turnover by promoting keratinization, which facilitates the smooth discharge of existing melanin from the skin. This action also regulates keratinocyte metabolism, corrects epidermal atypical hyperplasia, and promotes the increase of hyaline keratin particles, resulting in a thinner stratum corneum and an overall smoother, brighter skin surface.

Key findings

  1. 01 A 3% 4MSK formulation significantly improved skin lightness in both pigmented and non-pigmented areas of cheek skin and reduced desquamation in a human clinical study.
  2. 02 Concentrations from 1% to 5% 4MSK demonstrated a concentration-dependent protective effect against ultraviolet-induced pigmentation, with 3% showing efficacy after 15 days of continuous use.
  3. 03 In vitro studies using a human 3D epidermal equivalent demonstrated significant reduction in melanin amounts at a 0.05% concentration.
  4. 04 Cultured human melanocytes showed significantly reduced melanin contents when treated with 0.8-1.0 mM 4MSK in vitro.

Stability

Optimal stability is achieved within a pH range of 3.5 to 6.5. In acidic formulations, the system pH should remain above 5.0. It is crucial to avoid temperatures exceeding 60°C during the mixing process. For enhanced light resistance, the addition of 0.05%-0.50% BHT is recommended. For improved heat resistance, consider adding 0.05%-0.10% ammonium bisulfite or sodium pyrosulfite.

Conflicts

  • Acidic conditions below pH 5.0.
  • Its electrolytic nature may alter the texture of certain gel or cream formulations.
  • In ionic systems, an ion-resistant thickener is recommended for essences, and non-ionic emulsifiers for emulsions, to prevent stability issues.

Sensitization risk Low

Potassium Methoxysalicylate exhibits a favorable safety profile with minimal reported irritation at recommended concentrations. It received approval as an active skin lightening ingredient in Japan's quasi-drug category in 2003 and as a cosmetic raw material in China in 2007. A clinical study noted minor and temporary skin sensations (itching, prickling, burning) in a very small subset of subjects (1 out of 31 for each reaction), with no serious adverse events observed.
