Mechanism of Action
Phragmites Communis Extract operates through several intricate pathways to bolster skin vitality. It acts as a potent antioxidant, neutralizing free radicals to prevent oxidative damage. The extract actively inhibits melanin synthesis by suppressing tyrosinase activity and α-melanocyte-stimulating hormone-mediated melanin production, leading to brightening effects. It combats photoaging by downregulating matrix metalloprotease-1 (MMP-1) expression and elastase activity, enzymes crucial in collagen and elastin degradation. Furthermore, it upregulates hyaluronan synthase-2 (HAS-2) expression, thereby increasing endogenous hyaluronic acid content for enhanced hydration. Its anti-inflammatory properties soothe irritation, reduce tightness, and contribute to fortifying the skin's natural barrier and balancing the microbiome, promoting overall skin resilience.

Key findings
- 01 In human dermal fibroblasts and keratinocytes, a concentration range of 25-200 μg/mL demonstrated significant, concentration-dependent photoprotection. This involved downregulating MMP-1 expression and elastase activity in HDFs, and upregulating telomerase reverse transcriptase and hyaluronan synthase-2 expressions in HaCaT cells. It also suppressed α-MSH-mediated melanin production and tyrosinase activity.
- 02 At 25-50 μg/mL, the extract inhibited melanin production and cellular tyrosinase activity in B16F10 melanocytes stimulated with α-MSH. It suppressed mRNA expression of MMP-1 and reduced elastase activity in HDFs, while effectively suppressing UVB-mediated downregulation of HAS-2 gene expression in HaCaT keratinocytes. Notably, concentrations up to 200 μg/mL for 24 hours did not induce significant cytotoxicity.
