Mechanism of Action
This extract operates through several key pathways: it significantly reduces inflammation by inhibiting inflammatory responses, decreasing histamine release, and modulating various inflammatory cytokines (e.g., IL-1β, IL-4, IL-13, TNF-α) and chemokines via NF-κB, Akt, ERK1/2, and p38 MAPK pathways. It also boosts cellular antioxidant defenses by increasing heme oxygenase-1 (HO-1) expression via NRF2 nuclear translocation and upregulating other antioxidant genes like catalase (CAT) and glutathione peroxidase 1 (GPX1). Rich in active compounds such as rutin, pteryxin, chlorogenic acid, and coumarins, the extract further exhibits antiapoptotic effects, enhances re-epithelialization, suppresses collagen-degrading enzymes (MMP-1), and promotes collagen type I alpha 1 (COL1A1) synthesis.

Key findings
- 01 Peucedanum japonicum root extract demonstrated concentration-dependent inhibition of scratching behavior, significant reduction of inflammatory cells and mast cell infiltration, and decreased secretion of histamine and inflammatory cytokines/interleukins in both mice and human skin mast cells and macrophages, indicating efficacy against atopic dermatitis-like symptoms.
- 02 Chlorogenic acid, a major component of the extract, at 50 μg/mL, decreased intracellular reactive oxygen species (ROS), suppressed matrix metalloproteinase-1 (MMP-1) secretion, and enhanced collagen type I alpha 1 (COL1A1) expression in human dermal fibroblasts stimulated by TNF-α.
- 03 Peucedanum japonicum-derived exosome-like nanovesicles (PjELNs) exhibited robust anti-inflammatory effects in a contact dermatitis model, reducing ear swelling, histopathological damage, inflammatory cell infiltration, and the expression of inflammatory cytokines and chemokines (CXCL2, IL-1β, IL-4, IL-13, and TNF-α).

Stability
The extract's stability is bolstered by its natural content of phenolic compounds, including chlorogenic, neochlorogenic, and cryptochlorogenic acids, which are renowned for their antioxidant activity and capacity to mitigate oxidative stress. Extracts are commonly prepared using water and dissolved in methanol for analytical purposes.
Safety
Safety Profile
The Cosmetic Ingredient Review (CIR) Expert Panel has not formally reviewed Peucedanum Japonicum Extract. Acute oral toxicity tests in rats administered 5000 mg/kg of Peucedanum japonicum leaf extract (PJE) showed no signs of clinical toxicity or body weight changes over 15 days. In in vitro studies, some root extracts displayed mild cytotoxicity at the highest tested concentration (50 μg/mL) in human dermal fibroblasts, while leaf extracts exhibited no cytotoxic effects. It is generally classified with a low potential for skin, eye, or lung irritation and low non-reproductive organ system toxicity. Paula's Choice further indicates it as generally non-irritating.
