Mechanism of Action
This extract confers its benefits through several sophisticated molecular mechanisms. It notably elevates skin moisture levels. For skin repair, it stimulates the proliferation and sprout outgrowth of human epidermal keratinocytes and modestly enhances their migration, concurrently upregulating Type I and IV collagen synthesis and filaggrin expression via the AKT/MAPK pathway. Its anti-melanogenic action involves inhibiting melanin production and intracellular tyrosinase activity by downregulating Microphthalmia-Associated Transcription Factor (MITF) and regulating the MAPK signaling pathway. Additionally, due to its phenolic compounds and flavonoids, it exhibits antioxidant and anti-inflammatory properties, alongside traditional antimicrobial activity.

Key findings
- 01 A clinical study demonstrated that an extract doubled skin moisture levels immediately after the first application, with high hydration sustained for over three hours. Before application on Day 8, extract-treated skin exhibited more than 20% higher moisture levels compared to placebo controls, and the extract enhanced the moisturizing action of the placebo by 50% after Day 8 application.
- 02 In vitro studies at 50, 100, and 200 µg/mL showed Paederia foetida flower absolute (PFFA) increased human epidermal keratinocyte (HaCaT cell) proliferation and sprout outgrowth, modestly enhanced migration, elevated phosphorylation of ERK1/2, AKT, and p38 MAPK, and upregulated Type I and IV collagen synthesis and filaggrin expression, indicating promotion of skin wound and barrier repair.
- 03 In vitro research with Paederia foetida L. extract (PFE) at 50, 100, and 200 µg/mL significantly inhibited melanin content and intracellular tyrosinase activity in B16F10 murine melanoma cells, without cytotoxicity. PFE strongly inhibited the protein expression of tyrosinase-related proteins (TRP)-1, -2, tyrosinase, and microphthalmia-associated transcription factor (MITF) by regulating the MAPK signaling pathway.
