Science

Osthole exerts its effects through multiple cellular pathways. It significantly reduces inflammation by inhibiting NF-κB activation, modulating the production of pro-inflammatory cytokines (IL-1β, TNF-α, CCL2/MCP-1, CCL5/RANTES) and chemokines (IL-8), and downregulating TLR2 and COX-2 expression. It also blocks MAPK/p38 pathways and inhibits 5-lipoxygenase. For barrier integrity and wound healing, Osthole enhances ZO-3 expression and stimulates keratinocyte proliferation and migration. Its antipruritic action stems from stabilizing mast cell membranes and inhibiting histamine release, while also blocking substance P signaling. Furthermore, Osthole contributes to anti-photoaging by decreasing oxidative stress, exhibits hygroscopic properties for moisturizing, and demonstrates broad-spectrum antibacterial and antifungal activity. Key findings

Stability

Under U.S. law, cosmetic ingredients generally do not require pre-market FDA approval. While acute rodent toxicity studies for internal administration indicated an LD50 of approximately 710 mg/kg (intraperitoneal in mice) and subchronic oral administration in rats showed a NOAEL of less than 5 mg/kg/day with renal impairment at higher doses, these findings are for internal use and do not directly translate to topical cosmetic application.
