Mechanism of Action
Nerolidol disrupts bacterial cell membranes, leading to increased leakage of K+ ions and macromolecules, ultimately causing cell lysis, and interferes with genes regulating pathogen pathogenicity. In fungal infections, it causes alterations in hyphal morphology and membrane permeability, resulting in hyphal shrinkage and partial distortion. Additionally, it enhances transdermal delivery of other compounds and exhibits antioxidant and anti-inflammatory actions by inhibiting the release of proinflammatory cytokines and mediators, and preventing glial cell activation.

As a tertiary alcohol, Nerolidol generally resists oxidation better than primary or secondary alcohols. However, like other terpenes, it can undergo polymerization via oxidation under extreme conditions and oxidizes over time when exposed to air. For optimal stability, store in tightly closed containers in a cool, well-ventilated area, away from high temperatures, sparks, open flames, and sources of ignition. The pH is generally neutral.

Nerolidol is recognized as safe for intended uses in cosmetics and as a food flavoring agent by the U.S. FDA (21 CFR 172.515). Human maximization tests and repeated insult patch tests at 4% showed no positive reactions for skin sensitization. However, weak reactions were observed in two adjuvant tests in guinea pigs at 3% and 10%, and it is classified as a Skin sensitizer Category 1B (H317) and Eye irritant Category 2 (H319) under hazard classifications. It complies with EU Cosmetics Regulation 1223/2009 and is not one of the 26 mandatory declarable allergens.
