Mechanism of Action
This ingredient functions by elevating the polyamine pool, particularly during the telogen (resting) phase, which subsequently induces characteristics of the anagen (growth) phase, including pigmentation, proliferation of follicular keratinocytes, and upregulation of beta-catenin. It demonstrates anti-oxidative properties by diminishing intracellular reactive oxygen species (ROS) and exhibits anti-inflammatory effects through the reduction of pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6. Its molecular structure allows for penetration through the epidermal layer, reaching the dermis to exert its effects.

Key findings
- 01 In an in vitro study, N(1)-methylspermidine at a concentration of 0.5 µM significantly prolonged the anagen phase of hair follicles, reduced cell apoptosis, and stimulated the expression of epithelial stem cell-associated keratin K15. It also decreased lactate dehydrogenase activity (a marker of cell death) and intracellular reactive oxygen species production in cultured keratinocytes, while reducing pro-inflammatory cytokine (TNF-α, IL-1β, IL-6) gene and protein expression after lipopolysaccharide stimulation, indicating anti-oxidative and anti-inflammatory benefits.
- 02 A clinical trial involving 60 male and female subjects experiencing chronic telogen effluvium demonstrated that daily topical application of a lotion containing N1-methylspermidine (alpha-methylspermidine) significantly reduced hair loss and stimulated hair growth. After 3 months of treatment, improvements were observed across key parameters including hair density, hair shaft diameter, percentage of anagen hair, and Hair Mass Index, with statistically significant efficacy generally maintained for 3 months following the cessation of treatment.
