Mechanism of Action
Menadione operates through multiple mechanisms to enhance skin health. It actively promotes cellular regeneration, diminishes inflammatory responses, and may improve skin elasticity and hydration by stimulating microcirculation within the dermal layers. Its depigmenting action is attributed to a dose-dependent decrease in melanin synthesis within Mel-Ab cells, achieved via ERK activation which subsequently downregulates MITF and tyrosinase protein levels. Additionally, menadione can protect human skin keratinocytes from the inhibitory effects of certain factors by binding to and inhibiting specific protein tyrosine phosphatases (PTPs). At low concentrations, it is capable of generating intracellular reactive oxygen species (ROS).
