- 01 In vitro, an Alfalfa Polysaccharide Extract (APE) at 25-400 µg/mL increased HaCaT cell viability. Specifically, 100-200 µg/mL APE triggered cornified envelope formation and enhanced gene expression of filaggrin, involucrin, and claudin1. Anti-inflammatory effects were noted at 50, 100, and 200 µg/mL APE by decreasing LPS-induced proinflammatory cytokine production. In primary normal human dermal fibroblasts (NHDF), APE increased procollagen synthesis and reduced elastase activity and metalloproteinase-1 (MMP) secretion.
- 02 An in vitro Medicago sativa L. seeds extract at 0.25-10% significantly enhanced the viability and proliferation of skin cells (fibroblasts and keratinocytes), increasing fibroblast viability by up to 130% and keratinocyte viability by almost 160% at a 5% concentration. The extract also demonstrated notable antioxidant potential, effectively reducing intracellular reactive oxygen species.
