MEDICAGO SATIVA SEED EXTRACT.

Valuable CAS 84082-36-0 / SKIN CONDITIONING

Medicago Sativa Seed Extract, derived from alfalfa, is a multi-functional botanical ingredient recognized for its retinoid-like properties. It visibly reduces the signs of aging by boosting collagen and enhancing skin elasticity, while its rich antioxidant profile defends against environmental stressors. This extract promotes cellular regeneration, leading to a firmer, more radiant, and resilient complexion.

Antioxidant Skin-conditioning Anti-aging Firming Anti-inflammatory Brightening Oil-regulating Barrier-strengthening

Science

Operating with a retinoid-like action, Medicago Sativa Seed Extract stimulates dermal metabolism, leading to increased collagen type 1 synthesis and inhibition of metalloproteinase-1 (MMP-1) activity, key factors in combating wrinkles and improving skin firmness. It fosters cell regeneration to enhance skin vitality and elasticity, promoting a brighter, more youthful appearance. The extract is rich in galactomannans, vitamins (A, B, C, D, E, K), minerals, and potent antioxidants like chlorophyll, phenolic compounds, and flavonoids, which collectively combat free radicals, reduce oxidative stress, and mitigate pollutant damage. It strengthens the skin barrier by increasing epidermal cell differentiation and enhancing tight junction protein (claudin1) formation, while also exerting anti-inflammatory effects by reducing proinflammatory cytokines. Furthermore, it supports improved microcirculation to reduce puffiness and dark circles, regulates oil production due to B vitamins, and exhibits anti-melanogenic properties by inhibiting melanin synthesis.

Key findings

  1. 01 In vitro, an Alfalfa Polysaccharide Extract (APE) at 25-400 µg/mL increased HaCaT cell viability. Specifically, 100-200 µg/mL APE triggered cornified envelope formation and enhanced gene expression of filaggrin, involucrin, and claudin1. Anti-inflammatory effects were noted at 50, 100, and 200 µg/mL APE by decreasing LPS-induced proinflammatory cytokine production. In primary normal human dermal fibroblasts (NHDF), APE increased procollagen synthesis and reduced elastase activity and metalloproteinase-1 (MMP) secretion.
  2. 02 An in vitro Medicago sativa L. seeds extract at 0.25-10% significantly enhanced the viability and proliferation of skin cells (fibroblasts and keratinocytes), increasing fibroblast viability by up to 130% and keratinocyte viability by almost 160% at a 5% concentration. The extract also demonstrated notable antioxidant potential, effectively reducing intracellular reactive oxygen species.
  3. 03 An in vivo study observed significant wrinkle reduction after 28 days of topical treatment with a formulation containing Medicago Sativa Extract. The specific concentration of the extract in the final product was not detailed.

Sensitization risk Low

Medicago Sativa Seed Extract is generally considered safe for topical cosmetic use, exhibiting anti-aging benefits without reported irritation or adverse effects in topical applications. In vitro studies confirm low cytotoxicity for skin cells. It carries a low hazard score for concerns such as cancer, allergies, immunotoxicity, and developmental toxicity according to EWG. SkinSAFE classifies it as 'Irritant Free', 'Teen SAFE', 'Lip SAFE', 'Baby SAFE', and 'Eyelid SAFE'. While the FDA lists Medicago Sativa (Alfalfa) Extract as a cosmetic ingredient, pre-market safety testing for cosmetics is not mandated. Oral ingestion of large quantities of alfalfa seeds has been linked to lupus-like symptoms due to canavanine; however, this is not relevant to topical application.
