Macelignan employs multiple pathways to enhance skin. It safeguards keratinocytes against UVB-induced damage by suppressing MMP-9 and COX-2 expression, achieved through attenuation of MAPK and PI3K/Akt pathway activation. To brighten skin, it decreases melanin biosynthesis by downregulating the expression of tyrosinase, TRP-1, and TRP-2 proteins. It also counteracts photoaging by mitigating UV-induced MMP-1 expression via the suppression of reactive oxygen species-induced MAPK phosphorylation, while simultaneously boosting type I procollagen expression through TGF-β/Smad signaling. Additionally, macelignan is reported to activate adipogenesis in skin fatty cells, contributing to increased subcutaneous volume that helps to smooth wrinkles and expression lines. It also possesses inherent antioxidant and anti-inflammatory attributes.
