IONONE.

Valuable CAS 8013-90-9 / 79-77-6 / ASTRINGENT, PERFUMING

Ionone, encompassing its alpha and beta isomers, is a versatile ingredient recognized for its ability to bolster skin health beyond its aromatic properties. It actively supports epidermal barrier repair, stimulates cellular regeneration, and enhances the production of crucial skin components like collagen and hyaluronic acid, fostering a more resilient and youthful complexion.

Science

Alpha-ionone improves keratinocyte functions by activating the cAMP signaling pathway, leading to increased cell proliferation and migration. This process also enhances the production of hyaluronic acid (HA) and human β-defensin-2 (HBD-2), thereby accelerating epidermal barrier recovery. Beta-ionone acts as an antagonist to the glucocorticoid receptor (GR), which helps to mitigate glucocorticoid-induced suppression of collagen and hyaluronic acid synthesis in human dermal fibroblasts. Furthermore, alpha-ionone has been shown to increase collagen density, boost collagen gene expression, reduce matrix metalloproteinase genes, and suppress senescence markers in models of photoaging.

Key findings

  1. 01 Topical application of 0.1%-1% α-ionone was observed to accelerate epidermal barrier recovery in human skin after barrier disruption.
  2. 02 In vitro studies demonstrated that 10-50 µM α-ionone promoted human immortalized keratinocyte (HaCaT) cell proliferation and enhanced cell migration.
  3. 03 Up to 200 µM β-ionone showed no negative impact on human dermal fibroblast viability and attenuated dexamethasone-induced suppression of collagen and hyaluronic acid synthesis.

Stability

Ionones are soluble in most fixed oils, alcohol, and propylene glycol, but show very slight solubility or are insoluble in water and glycerin. Alpha-ionone is light sensitive and requires storage in a cool, dry, tightly sealed container. Beta-ionone should avoid contact with strong Lewis or mineral acids, halogens, and free radical initiators. A pH of 7.0 has been successfully utilized in hydrogel formulations for alpha-ionone.

  • mineral acids
  • halogens
  • free radical initiators

Safety

CIR Status
Safe with restrictions
Sensitization risk Moderate

The Cosmetic Ingredient Review (CIR) Expert Panel has deemed Alpha-Isomethyl Ionone safe for cosmetic use at regulated concentrations, often deferring to the IFRA program for fragrance ingredients. The Scientific Committee on Consumer Safety (SCCS) of the EU Commission has approved its use with specific restrictions. While the FDA has approved Alpha-Isomethyl Ionone as a food flavoring agent, the International Fragrance Association (IFRA) Standard restricts its use in fragrances due to potential sensitization. Under EU regulations, Ionone (including Alpha-Isomethyl Ionone) must be declared on product labels if its concentration surpasses 0.001% in leave-on products and 0.01% in rinse-off formulations, owing to its classification as a potential allergen.
