Mechanism of Action
This extract operates through several key mechanisms: it scavenges reactive oxygen species (ROS) to protect cellular membranes from oxidative damage, thereby conferring antioxidant effects. It inhibits tyrosinase, a crucial enzyme in melanin synthesis, to mitigate hyperpigmentation. Furthermore, its active compound, 1-O-acetylbritannilactone, demonstrates significant anti-inflammatory actions by inhibiting human neutrophil elastase (HNE), suppressing the production of inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2), and downregulating the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2. The extract also exhibits antibacterial activity.

Key findings
- 01 Suppressed photohemolysis in human erythrocytes in a concentration-dependent manner, indicating cellular protective and antioxidant effects.
- 02 Its isolated compound, 1-O-acetylbritannilactone, significantly inhibited human neutrophil elastase (HNE) with an IC50 of 3.2 ± 0.3 µM, demonstrating potent anti-inflammatory effects.
- 03 At 500 µg/ml in cell culture, the extract inhibited tyrosinase activity by 29.33% to 41.74% (when fermented), suggesting antimelanogenic/skin-whitening effects with maintained high cell viability (100.26% for fermented extract, 92.15% for non-fermented).

Safety Profile
The Cosmetic Ingredient Review (CIR) Expert Panel has not formally reviewed this ingredient for cosmetic safety. However, independent assessments, such as EWG Skin Deep®, rate Inula Britannica Flower Extract with 'LOW' concerns for Cancer, Allergies & Immunotoxicity, and Developmental and Reproductive Toxicity. It's important to note that the plant's addition to the U.S. Federal Noxious Weed List in 2010 relates to agricultural and environmental concerns, not direct safety for topical cosmetic application of the extract.
