Mechanism of Action
HU-MSCCM operates through a multifaceted action, delivering a complex blend of growth factors, including EGF, bFGF, TGF-β, PDGF, HGF, Collagen type 1, and GDF-11. These biomolecules stimulate the proliferation, differentiation, and regeneration of dermal fibroblasts and promote keratinocyte migration. Its anti-inflammatory effects are achieved by modulating the immune response, specifically inhibiting pro-inflammatory type 2 cytokines and chemokines (e.g., TARC, TNF-α, IL-6, IL-4, IL-13). The ingredient strengthens the skin barrier, accelerates wound healing through enhanced angiogenesis, cell migration, fibroplasia, and maturation. For anti-aging, it reduces UV damage and mitigates photoaging by lowering apoptosis, decreasing reactive oxygen species (ROS) generation, enhancing cell motility, and inhibiting UV-induced suppression of SOD and GSH-Px activities while blocking malondialdehyde upregulation. It also inhibits melanogenesis by regulating microphthalmia-associated transcription factor (MITF) expression via the ERK signaling pathway, leading to proteasomal degradation of MITF. Furthermore, it boosts the production of Collagen type 1, Collagen type 3, Fibronectin, and Elastin in Human Dermal Fibroblasts (HDFs) and reduces Matrix Metalloproteinase-1 (MMP-1) expression.

Clinical Evidence
High confidenceN/A
Key findings
- 01 Topical application significantly improved skin barrier function and reduced inflammatory responsiveness, yielding beneficial effects on sensitive skin with no severe adverse events reported.
- 02 A 4-week application to mild Atopic Dermatitis lesions strengthened the skin barrier, increasing Corneometer levels and decreasing transepidermal water loss (TEWL).
- 03 Demonstrated anti-wrinkle effects and a significant increase in dermal density in women.
- 04 In a cream formulation, it promoted primary wound healing regeneration on incision skin injury in vivo, decreasing inflammation, fostering angiogenesis, and accelerating fibroplasia and maturation phases.
- 05 Effective in preventing and treating skin aging by stimulating fibroblast proliferation, strongly inhibiting melanin production, and increasing skin thickness, Ki-67-positive cells, and collagen deposition in aged models.

Formulation
Stability
As a complex mixture of biomolecules, including proteins and growth factors, this ingredient requires careful handling. Cryo-dehydrated powder, when mixed with a cosmetic base, has shown stability for approximately one week under refrigerated conditions.
Safety
Safety Profile
Non-clinical safety evaluations indicate this conditioned medium is a safe substance. Clinical studies reported no severe adverse events following topical application on sensitive skin or for mild Atopic Dermatitis. It is important to note that FDA warnings regarding unapproved umbilical cord *products* (cellular/tissue products) for therapeutic use do not directly apply to conditioned media used as a cosmetic ingredient.
