Mechanism of Action
This extract exerts its effects by modulating cytokine expression and NF-κB signaling, thereby reducing key inflammatory mediators like IL-6 and IL-17C, as well as allergic mediators such as TSLP, CCL26, and MMP-9. Its rich tannin content offers astringent action by precipitating surface proteins, which helps to reduce excess sebum, minimize redness, and strengthen the skin barrier. The extract also functions as a powerful antioxidant, enhancing cellular defenses by reducing reactive oxygen species (ROS) and inhibiting enzymes like elastase and collagenase, offering protection against UV-induced damage and pollution. Additionally, it exhibits vasoconstrictive properties and helps maintain skin barrier integrity through regulating tight junction proteins.

Key findings
- 01 Inhibited the release of mediators involved in skin autoimmunity (IL-6, IL-17C) and allergy (TSLP, IL-6, CCL26, MMP-9) in human keratinocytes, demonstrating anti-inflammatory and anti-allergic potential.
- 02 Leaf and bark extracts significantly reduced hydrogen peroxide-induced reactive oxygen species (ROS) accumulation in human keratinocytes and dermal fibroblasts, highlighting antioxidant capabilities.
- 03 Showed protective effects against fine dust-induced damage in skin cells, suggesting an anti-pollution benefit.
- 04 Applied in a cream, it showed no reported adverse effects and offered protection to skin damaged by UVB light.
- 05 Multiple HRIPTs and patch tests reported no irritation or sensitization, affirming a low irritation potential even at higher concentrations.
- 06 Demonstrated significant reduction (56% to 93%) of biofilm formation for various Candida species, E. coli, K. pneumoniae, S. aureus, and S. mutans, indicating broad-spectrum antibacterial properties.

The Formula
Formulation
Stability
The chemical profile and stability can vary based on the extraction solvent and method (e.g., hydroglycolic, hydroalcoholic, or glycerin/water-based extracts). Care should be taken to avoid alcohol-containing distillates (which can contain up to 14% added alcohol) in formulations targeting sensitive or dry skin, as these can be drying or damaging.
Conflicts
- High concentrations of the extract may lead to irritation and dryness in some individuals.
- Distillates containing alcohol should be avoided or carefully considered, especially for sensitive or dry skin types, due to potential drying or damaging effects.
- Cumulative high concentrations of shared botanical constituents (e.g., geraniol, oxidation products of linalool) when combined with other botanical ingredients in a formulation may increase the risk of sensitization or adverse reactions.
Safety
Safety Profile
The CIR Panel concluded that Hamamelis virginiana-derived ingredients are safe for cosmetic use when formulated to be non-irritating and non-sensitizing. The highest reported leave-on concentration for Hamamelis Virginiana (Witch Hazel) Extract in cosmetics is 1.8%. However, Hamamelis Virginiana (Witch Hazel) Water has been used safely up to 43% in other skincare preparations, and a 10% Hamamelis Virginiana Bark/Twig Extract showed no adverse effects on UVB-damaged skin. The FDA has approved witch hazel for use in over-the-counter (OTC) formulations for minor skin irritations and hemorrhoidal/vaginal symptoms. No SCCS review was found.
