Mechanism of Action
Glutathione primarily modulates skin pigmentation by inhibiting the tyrosinase enzyme, which is critical for melanin biosynthesis, and shifting melanogenesis from darker eumelanin towards lighter phaeomelanin. As a potent antioxidant, it neutralizes free radicals and mitigates oxidative stress induced by UV exposure and environmental aggressors, thereby preventing skin aging and damage. It also supports collagen synthesis, enhancing skin elasticity, reducing wrinkle appearance, and promoting tissue repair. Furthermore, glutathione can improve skin hydration by decreasing trans-epidermal water loss (TEWL), inactivates tyrosinase by binding copper ions, interferes with the intracellular trafficking of melanogenic enzymes, and interacts with dopamine to reduce melanin formation while protecting pigment cells.

Key findings
- 01 Topical oxidized glutathione brightened skin color in sun-exposed areas, improved skin wrinkle, elasticity, and UV spots.
- 02 Topical glutathione lotion significantly reduced melanin indices, improved skin moisture and texture, and reduced melasma severity.
- 03 S-acyl glutathione cream significantly reduced ultraviolet-induced erythema.
- 04 Skincare products containing 0.5% glutathione were more effective than 0.1% in improving skin brightness and reducing hyperpigmented spots within eight weeks.
- 05 Provides antioxidant properties and contributes to a radiant glow.
- 06 Effectively disrupts tyrosinase activity for potent skin brightening.

Dusting Analysis
Clinical evidence demonstrates dose-dependent efficacy for topical glutathione. While some benefits for skin brightness were observed at 0.1%, significantly superior results for hyperpigmentation reduction and overall skin improvement are consistently seen at 0.5% and 2.0%. Concentrations below 0.5% may offer suboptimal active benefits for comprehensive brightening and anti-aging outcomes.

Stability
Glutathione is highly unstable in water-based formulations at room temperature. A low pH environment may also negatively impact its stability, requiring careful formulation strategies.
Conflicts
- Vitamin B12
- AHA/BHA products
Safety
Safety Profile
While the FDA in the Philippines warns against injectable glutathione due to systemic toxic effects, and oral glutathione efficacy for skin is still being assessed, topical glutathione is generally well-tolerated. Some mild localized reactions, such as transient erythema or a brief tingling sensation, have been reported in a small percentage of participants. Instances of acne worsening were noted in products containing 0.1-0.5% reduced glutathione. Overall safety data for topical application remains limited and warrants further extensive exploration.
