Mechanism of Action
This extract functions as a potent antioxidant, primarily through garcinol and anthocyanins, by neutralizing free radicals (e.g., DPPH, H2O2) and influencing arachidonic acid metabolism and nitric oxide (NO) radical synthesis to maintain oxidative balance. Its anti-inflammatory properties stem from suppressing NFκB activation, inhibiting iNOS synthesis, and diminishing the production of extracellular signal-regulated kinase 1/2, cyclooxygenase-2, and prostaglandins. Garcinia Indica also inhibits 5-lipoxygenase, reducing inflammatory leukotriene production. Beyond these actions, it supports cell proliferation and migration, aiding in wound healing, and exhibits anti-allergenic effects by decreasing histamine release. Certain components, like Kokum Butter, provide significant emollient benefits, while the extract also demonstrates antibacterial and antiviral activity.

Key findings
- 01 In vitro, Garcinia Indica fruit extract demonstrated a 57% inhibitory effect on DPPH activity and 65% for H2O2 radical scavenging, indicating significant antioxidant capacity.
- 02 A standardized SPF formulation containing 2.5% Garcinol-enriched Ethyl Acetate Extract (TEAE) synergistically achieved an SPF of 14 when combined with 3% Octyl Methoxycinnamate (OMC), a notable increase compared to OMC alone (8.05) or TEAE alone (3.68).
- 03 A base formulation supplemented with 1% Kokum butter exhibited 96.00% scratch healing ability on mice fibroblast cell lines in vitro, indicative of enhanced cell migration and proliferation crucial for skin repair.
- 04 A cream formulation containing 1% Garcinol-enriched Ethyl Acetate Extract (TEAE) maintained 100.87% cell viability on 3T3 mice fibroblast cells, confirming its non-cytotoxic profile at this concentration.
- 05 Methanolic extracts of Garcinia indica did not cause 50% cell death on mouse embryo 3T3 fibroblast cells up to 400 µg/mL, suggesting non-cytotoxicity for normal fibroblast cells.

Stability
Garcinol, a key active, is fat-soluble, while anthocyanins, another beneficial component, are water-soluble pigments. The anthocyanin colorant from Kokum shows high stability within a pH range of 1.0–3.0 but becomes colorless at pH 4.5 and blue above pH 4.5. Highly acidic fruit rind extracts (e.g., pH 1-2) are generally not suitable for cosmetic formulations and may require washing to achieve a more suitable pH of around 6.
Synergies
- Octyl Methoxycinnamate (OMC) for enhanced SPF.
- Kokum butter to ameliorate potential cytotoxicity of Garcinol-enriched Ethyl Acetate Extract (TEAE) at higher concentrations.
Conflicts
- Highly acidic pH (1-2) for direct cosmetic formulation without adjustment.
- High concentrations of certain enriched extracts (e.g., TEAE) may reduce cell viability in vitro if not properly formulated or combined with emollients like Kokum butter.
Safety
Safety Profile
Garcinia Indica is recognized as safe for cosmetic and personal care products by leading regulatory bodies including CIR, FDA, and EU Cosmetics Regulation. While some Garcinia species (like G. cambogia) have raised concerns regarding hepatotoxicity as food supplements, no direct association with G. indica for acute liver injury has been reported in cosmetic use. Cosmetic formulations containing ethyl acetate extract and Kokum butter have been assessed as practically non-irritant in in-vitro studies on mice fibroblast cells. However, in vitro studies indicated a Garcinol-enriched Ethyl Acetate Extract (TEAE) had an IC50 of 63.32 µg/ml and a 2.5% TEAE cream formulation reduced cell viability significantly at 1 mg/mL (10.44%), an effect which was mitigated when fortified with Kokum butter.
