Mechanism of Action
The extract's efficacy stems from its rich profile of polysaccharides, triterpenes, peptides, polyphenols, and flavonoids, which collectively provide robust antioxidant defense against free radicals and oxidative stress. It functions as an anti-inflammatory agent, effectively reducing irritation and soothing the skin. For anti-aging, it promotes natural collagen production, improving skin elasticity and firmness while positively influencing collagen fiber organization. It also interrupts tyrosinase, the enzyme responsible for melanin synthesis, leading to a visible reduction in discoloration and counteracting UVB-induced pigmentation. Additionally, it strengthens the skin barrier, preventing moisture loss and protecting against environmental stressors, and supports skin regeneration and hydration.

Key findings
- 01 Extracts, at concentrations up to 2 mg/mL, demonstrated no overt toxicity to human keratinocytes (HaCaT) and exhibited superior scavenging ability on ABTS and DPPH radicals. A hot water crude polysaccharide extract (HWCP) showed potent inhibition of tyrosinase and skin extracellular matrix (ECM) degradation enzymes.
- 02 A proprietary reishi extract (RE) significantly enhanced skin firmness and collagen organization by stimulating dermal fibroblasts, attributed to its antioxidant capacity, increased synthesis of structural proteins, and influence on collagen fiber organization.

Stability
The extract's bioactivities are preserved in cosmeceutical formulations at a pH of 4.6. Optimal extraction temperatures vary: 60°C for hot air drying best preserves ganoderic acid, while freeze-drying at -50°C is optimal for antioxidant and antidiabetic activities. High heat (80°C or higher) or extended drying periods should be avoided as they can degrade active ingredients.
Safety
Safety Profile
Ganoderma lucidum extract exhibits an excellent safety profile and is generally considered non-irritating. Acute and subacute toxicity tests in mice showed no serious or lethal effects at doses exceeding 5,000 mg/kg. Most hydroalcoholic extracts do not show cytotoxic effects on normal cells at high proliferation rates. While rare, some individuals may experience mild skin irritation or allergic reactions.
