Mechanism of Action
This extract leverages a complex array of bioactive compounds, including phenolic compounds, triterpenes, and polysaccharides. Its action involves significant antioxidant activity, neutralizing free radicals and mitigating oxidative stress. It effectively inhibits tyrosinase, an enzyme crucial for melanin production, thereby contributing to skin brightening. Furthermore, it demonstrates anti-collagenase and anti-elastase activities, preventing the enzymatic degradation of collagen and elastin to support skin's structural integrity. Immunomodulatory and antimicrobial properties also contribute to a resilient skin barrier.

Key findings
- 01 In in vitro cellular studies, methanol and hot water extracts from Ganoderma applanatum fruiting bodies demonstrated good anti-tyrosinase (skin whitening), good anti-collagenase, and moderate anti-elastase (anti-wrinkle) activities. Specifically, cellular melanin synthesis in B16-F10 melanoma cells was inhibited by over 50% at a concentration of 2.0 mg/mL.
- 02 An isolated exo-biopolymer produced by Ganoderma applanatum, composed of 58.9% carbohydrate and 17.1% protein, increased phosphatase activity of macrophages by 0.7-fold at 200 microg/mL. This exo-biopolymer also exhibited antitumor effects and enhanced natural killer (NK) cell activity in mice at a dosage of 40 mg/kg BW.

Stability
While direct stability data for the extract in cosmetic formulations is not available, studies on mycelial growth indicate optimal growth and exo-polymer production are typically observed in a pH range of 5-6, suggesting potential stability across a broad pH spectrum.
Safety
Safety Profile
The Cosmetic Ingredient Review (CIR) Expert Panel has not specifically evaluated Ganoderma applanatum mycelium extract. However, microbial polysaccharide gums are generally considered safe for cosmetic use. The FDA acknowledges mushroom mycelium as suitable for food use when correctly labeled, providing an indirect indication of its general safety profile, though this is not a direct assessment for topical cosmetic application. Irritancy and comedogenicity data are currently unknown.
