Mechanism of Action
This extract operates through a multifaceted mechanism rooted in its rich composition of polyphenolic compounds. These compounds exert potent antioxidant activity, effectively neutralizing free radicals and diminishing the visible effects of inflammatory responses often triggered by sun exposure. It provides significant anti-inflammatory and soothing benefits by reducing the expression of inflammatory markers like COX-2 and iNOS in impaired skin cells. Furthermore, the extract demonstrates antimicrobial efficacy against bacteria such as Propionibacterium acnes, making it a promising agent for managing acne-prone skin. Specific phenolic compounds isolated from the leaves also contribute to anti-photoaging by increasing the viability of human epidermal keratinocytes.

Key findings
- 01 Isolated phenolic compounds (protocatechuic acid, epicatechin, and kaempferol) derived from Entada phaseoloides leaves, at a concentration of 100 μM, significantly increased human epidermal keratinocyte (HaCaT) cell viability in UVB-irradiated conditions and suppressed COX-2 and iNOS inflammation gene expression, demonstrating anti-photoaging effects.
- 02 A hydroalcoholic extract from Entada phaseoloides seeds, at 100 mg/ml, exhibited notable antimicrobial activity against Propionibacterium acnes (18±0.57mm zone of inhibition) and was observed to reduce acne severity in rat models.
- 03 Skincare formulations incorporating a bark/seed extract blend of Entada phaseoloides typically utilize concentrations ranging from 1.5% to 4%.

Stability
Entada phaseoloides extracts exhibit robust stability across a broad range of pH values, as well as in the presence of various minerals and yeast cells. Stability is also maintained following exposure to ultraviolet irradiation of solutions. The extract can be prepared using organic solvents like acetone and methanol, and contains components with diverse solubilities in aqueous, chloroform, and petroleum ether fractions.
Safety
Safety Profile
Official safety assessments by regulatory bodies such as CIR, SCCS, or FDA are not yet available for Entada Phaseoloides Leaf Extract. However, it is specifically noted that the leaves of Entada phaseoloides are free from the toxic saponins found in other plant parts (seeds, bark), suggesting a more favorable safety profile for the leaf extract. In vitro studies showed that most isolated phenolic compounds from the leaf extract demonstrated no cytotoxicity in human epidermal keratinocyte (HaCaT) cells at 100 μM. While crude seed extracts have indicated low acute oral toxicity (LD50 > 5000 mg/kg in rats), other plant parts have been associated with general toxicity, molluscicidal activity, eye irritation, and detectable genotoxicity. These concerns are not attributed to the leaf extract.
