Mechanism of Action
Emodin operates through several pathways to improve skin health. It inhibits the proliferation and induces apoptosis of sebocytes, thereby controlling sebum production. It significantly reduces sebaceous lipogenesis by decreasing lipid synthesis and suppressing key lipogenic transcription factors (PPARγ, LXR α/β, SREBP-1), primarily by inhibiting the phosphoinositide-3-kinase (PI3K)/Akt/forkhead box protein O1 (FoxO1) signaling pathway. Furthermore, Emodin demonstrates anti-inflammatory effects by lowering pro-inflammatory cytokine levels (IL-6, IL-8, TNF-α) and suppressing the NLRP3 inflammasome pathway, partly through inhibiting TLR4 expression and reducing reactive oxygen species (ROS). It also functions as an antioxidant by scavenging ROS and exhibits antibacterial efficacy against Cutibacterium acnes.

Emodin exhibits poor water solubility (0.07 mg/mL) and is best incorporated into formulations via the oil phase or with the aid of suitable solubilizers. It should be added under controlled temperatures below 60 °C. Solutions in water, DMSO, 95% ethanol, or acetone maintain stability for 24 hours under normal laboratory conditions; however, prolonged exposure to light may affect its stability.

High-dose oral ingestion of anthraquinone derivatives, such as aloe-emodin, has raised concerns regarding potential liver and kidney toxicity, which is not applicable to topical cosmetic use.
