Mechanism of Action
Operating as a senomorphic peptide (also known as Pep 14 or OS-01), Decapeptide-52 intervenes in the cellular senescence pathway. Its action specifically reduces the mRNA expression of CDK2AP1, which subsequently activates CDK2, likely through the stabilization of protein phosphatase 2A (PP2A). This activation initiates cell cycle arrest, facilitating crucial DNA repair processes. By doing so, it effectively prevents pre-senescent cells from progressing into late-senescent cells and accumulating, ultimately decreasing the number of cells that secrete pro-aging factors (SASP).

Key findings
- 01 A significant study utilizing human skin models at a 0.01% w/v concentration of Decapeptide-52 demonstrated marked improvements in histological scores. It effectively reduced mRNA levels associated with senescence and inflammation, while notably increasing both keratin and collagen expression. Furthermore, it enhanced cell proliferation and was shown to safely decrease the skin's biological age, promote epidermal thickening, and improve various markers of skin health. The research indicated that Decapeptide-52 can outperform topical Retinol in certain aspects by precisely suppressing senescence through the prevention of late senescent cell accumulation.

Stability
Decapeptide-52 powder maintains stability for 24 months when stored between -20℃ and -15℃ in a freezer, provided it is protected from light and kept airproofed when not in use.
Safety
Safety Profile
While a clinical study reported Decapeptide-52 as safely reducing the biological age of skin in models, there are currently no specific safety assessments from major regulatory bodies such as CIR or SCCS. The FDA has not listed Decapeptide-52 among peptides posing significant safety risks as bulk drug substances for compounding, however, this does not constitute a formal approval or specific safety status for cosmetic use.
