Mechanism of Action
This synthetic peptide functions as a competitive antagonist to alpha-melanocyte-stimulating hormone (α-MSH), effectively binding to the MC1-R receptor on melanocytes. By blocking α-MSH activation, D-NONAPEPTIDE-1 disrupts the signaling cascade that leads to increased tyrosinase activity and melanin production. It systematically downregulates the expression of key enzymes like tyrosinase, TRP1, and TRP2, as well as MITF, and slows melanosome transfer, resulting in an approximate 33% reduction in overall melanin synthesis without inducing cytotoxicity or interfering with normal melanocyte function.

Key findings
- 01 A facial gel containing 0.0001% pure peptide significantly reversed hyperpigmentation within 28 days, leading to a 6.42% increase in skin lightness (△L) and a 5.39% improvement in comprehensive skin state (△E) by day 56.
- 02 A 0.0005% concentration of the peptide in a facial gel also demonstrated significant reversal of hyperpigmentation within 28 days.
- 03 An estimated skin lightening effect of at least 33% was observed, with continued improvement over time. A prospective double-blinded, randomized controlled pilot study lasting eight months showed observable amelioration in melasma severity scores and mean melanin index.
