Mechanism of Action
This brown algae extract works through multiple pathways to support skin health. Its hygroscopic nature enables it to act as a humectant, drawing moisture into the skin for a smooth, supple texture. The extract exhibits robust antioxidant activity by directly scavenging hydroxyl (OH) and nitric oxide (NO) radicals and reducing overall intracellular Reactive Oxygen Species (ROS) production, offering crucial photoprotection and contributing to anti-aging effects. Furthermore, it demonstrates potent anti-inflammatory properties by inhibiting key inflammatory mediators such as nitric oxide, IL-1α, IL-6, cyclooxygenase-2, and inducible NO synthase gene expression, effectively blocking toxicity in UV-exposed skin cells. Beyond this, it possesses anti-tyrosinase and anti-hydrolytic enzyme activities, counteracting collagenase and hyaluronidase to help mitigate uneven pigmentation and the formation of fine lines and wrinkles.

Key findings
- 01 Scavenged OH radicals in C. amentacea extracts (DMSO and ethanolic) with an EC50 ranging from 392 to 454 µg/mL.
- 02 Scavenged NO radicals in C. amentacea extracts (DMSO and ethanolic) with an EC50 ranging from 546 to 1293 µg/mL.
- 03 Demonstrated high radical scavenging potential in DPPH assay, achieving over 90% at highest concentrations, with EC50 values of 0.34 µg/mL for DMSO extract and 205.1 µg/mL for ethanolic extract.
- 04 Exhibited strong reducing activity, approximately 90% for DMSO extract and 75% for ethanolic extract at their highest tested concentrations, with EC50 values of 113.9 µg/mL and 643.5 µg/mL respectively.
- 05 Showed strong anti-inflammatory effects by inhibiting LPS-induced inflammatory mediators (NO production, IL-1α, IL-6, cyclooxygenase-2, inducible NO synthase gene expression) while demonstrating very low toxicity on RAW 264.7 macrophages and L929 fibroblasts.

Formulation
Stability
Polyphenols within algae extracts can exhibit thermal instability, with observed losses of 15-30% above 60°C. They are also susceptible to photo-oxidation, with 10-50% degradation upon UV-light exposure. Therefore, mild processing conditions and protection from high heat and UV light during storage and formulation are recommended to maintain ingredient integrity.
Safety
Safety Profile
The Cosmetic Ingredient Review (CIR) Expert Panel has concluded that existing data are insufficient to determine the safety of Cystoseira Amentacea/Caespitosa/Branchycarpa Extract for its intended use in cosmetic formulations. However, in vitro studies have indicated a very low grade of toxicity on RAW 264.7 macrophages and L929 fibroblasts. Due to its potent antioxidant activity and low observed toxicity in these studies, it has been suggested as safe for cosmeceutical applications. The ingredient is also categorized by SkinSAFE as '100% Free' of common allergens, Teen Safe, Baby Safe, and Eyelid Safe, collectively suggesting a low allergenic potential.
