Mechanism of Action
Coleus Forskohlii Root Extract exerts its effects through its key active compound, forskolin, which stimulates adenylate cyclase. This activation leads to an increase in intracellular cyclic AMP (cAMP) levels, a vital signaling molecule. Elevated cAMP can stimulate lipolysis (fat breakdown), help normalize disturbed cellular turnover, and induce the production of UV-protective eumelanin in melanocytes. Beyond these actions, the extract also demonstrates antioxidant, soothing, anti-inflammatory, and antimicrobial properties against common skin pathogens such as Propionibacterium acnes, Staphylococcus aureus, and Staphylococcus epidermidis.

Key findings
- 01 As a skin conditioning agent in topical formulations, concentrations of the extract between 0.1% and 0.5% are recommended.
- 02 Anti-cellulite formulations demonstrate benefits with a 95% pure forskolin content ranging from 1% to 3%.
- 03 Topical application of the extract has been shown in in-vitro and murine in-vivo studies to stimulate the production of UV-protective eumelanin and offer protection to skin exposed to UV light.

Formulation
Stability
Forskolin, the primary active, exhibits optimal stability within a pH range of 3.5 to 6.5. Its stability diminishes notably outside this range, with pseudo-first-order degradation kinetics observed between pH 6.5 and 8.5. The extract is available in both water-soluble and oil-soluble formats.
Synergies
- Incorporating 0.01% to 0.1% Cosmoperine can significantly enhance the transdermal penetration of the extract.
Safety
Safety Profile
The Cosmetic Ingredient Review (CIR) has not specifically assessed Coleus Forskohlii Root Extract for topical use. While dermal toxicity is listed as 'Not determined' by some sources, the root extract is generally not considered problematic for topical skin application. General safety information from oral intake studies, which is not directly transferable to topical concentrations, indicates no major adverse events at daily doses up to 1000 mg of extract. For oral consumption, caution is advised for individuals with hypotension or those on blood thinners due to potential interactions.
