Mechanism of Action
Chlorogenic Acids function as powerful antioxidants, actively scavenging various free radicals like hydroxyl, superoxide, and peroxylnitrite, thereby reducing oxidative stress induced by UV exposure. They exert anti-inflammatory effects by decreasing key markers such as IL-1β, TNF-α, COX-2, and nitric oxide. These compounds also support skin regeneration and anti-aging pathways by stimulating procollagen type I synthesis, upregulating essential skin barrier genes (filaggrin, involucrin, envoplakin), and inhibiting matrix metalloproteinases (MMPs) responsible for collagen degradation. Furthermore, they contribute to photoprotection by absorbing UV radiation and can act as an anti-acne agent through lipogenesis reduction and inflammatory cytokine downregulation.

- 01 In UV-exposed human skin fibroblast cells, Chlorogenic Acid demonstrated significant activity at specific concentrations. At 6.25 µg/mL, it notably reduced inflammatory markers nitric oxide (NO), cyclooxygenase-2 (COX-2), and interleukin-6 (IL-6), while also maximizing cell viability. A higher concentration of 25 µg/mL was effective in significantly downregulating genes associated with collagen degradation (MMP-1) and apoptosis (CASP-3, CASP-8). This concentration also boosted antioxidant defense (GPX-1) and extracellular matrix integrity (TIMP-1, FGF-2), concurrently decreasing reactive oxygen species (ROS), IL-1β, TNF-α, and cellular apoptosis/necrosis. Concentrations as high as 100 µg/mL showed reduced cell viability, indicating an optimal concentration range for beneficial effects in vitro.
