Mechanism of Action
This extract functions as a robust antioxidant by effectively scavenging free radicals and inhibiting the production of reactive oxygen species within skin keratinocytes. Its anti-inflammatory capabilities stem from modulating key signaling pathways, including PI3K/Akt and MAPK, and suppressing the expression of adhesion molecules like VCAM-1 and ICAM-1. It also reduces pro-inflammatory mediators such as TRPV-1 and MMP-1, which are implicated in inflammatory skin responses. Furthermore, the extract promotes anti-aging effects by increasing type-1 procollagen synthesis in dermal fibroblasts and providing protection against UV-induced damage.

Key findings
- 01 Demonstrated significant anti-inflammatory properties in human umbilical vein endothelial cells (HUVECs). It substantially reduced adhesion molecules VCAM-1 and ICAM-1 levels and attenuated PI3K/Akt signaling in LPS-treated HUVECs, with no observed cytotoxicity.
- 02 Provided photoprotective and anti-aging benefits. In human keratinocytes (HaCaT), it inhibited UVB-induced IL-6, IL-8, and PGE2 production by 25.6%, 5.3%, and 70.2% respectively. It also inhibited MMP-1 elevation by 20.0% and TRPV-1 by 41.9%. In human dermal fibroblasts (HDF), this concentration inhibited MMP-1 elevation and enhanced type-1 procollagen synthesis.
- 03 Exhibited potent antioxidant activity, demonstrating 92.36% free radical scavenging activity.

Safety Profile
The Cosmetic Ingredient Review (CIR) has not yet assessed Aralia Elata Leaf Extract for topical cosmetic use. In vitro studies on human umbilical vein endothelial cells (HUVECs) indicated no cytotoxicity at concentrations up to 5 μg/mL. While oral toxicity data in rats suggested a low acute and sub-chronic toxicity profile at tested dosages, this information does not directly establish a maximum safe topical concentration or confirm lack of sensitization in human skin.
