Mechanism of Action
Aralia Cordata Extract exerts its protective effects primarily by modulating inflammatory pathways and neutralizing oxidative stress. It inhibits key inflammatory signaling molecules suchs as NF-κB and MAPKs, reduces the production of pro-inflammatory mediators like COX-2 and PGE(2), and suppresses caspase-3 activity and histamine release. Concurrently, its rich composition of phenolic compounds, carotenoids, and anthocyanins enables it to scavenge free radicals and inhibit reactive oxygen species (ROS) generation, thereby mitigating oxidative damage to skin cells.

Key findings
- 01 Methanol extracts of Aralia cordata exhibited anti-inflammatory effects through inhibition of NF-κB and MAPKs signaling in LPS-stimulated RAW264.7 cells.
- 02 A constituent, 7-oxosandaracopimaric acid, inhibited COX-1 by 22.82 ± 1.94% and COX-2 by 15.86 ± 1.35% at 10 mM.
- 03 The constituent 7-oxosandaracopimaric acid significantly inhibited compound 48/80-induced histamine release from rat mast cells at 3.0 mM.
- 04 In cultured rat cortical neurons, the extract at 5-20 µg/mL inhibited hydrogen peroxide (H2O2)-induced apoptotic neuronal death, elevation of intracellular calcium concentration, and generation of reactive oxygen species (ROS).

Safety
Safety Profile
The Cosmetic Ingredient Review (CIR) has not evaluated Aralia Cordata Extract. While its root has a history of traditional use in Japan, Korea, and China without reported safety issues, an in vitro study observed cytotoxicity in cell lines with Aralia cordata root extracts. Furthermore, research on a related species, Aralia elata, indicated potential concentration-dependent toxicity, where cell viability significantly decreased at 10 μg/mL compared to 5 μg/mL on HUVECs.
