Mechanism of Action
Tetrapeptide-30 operates through a multifaceted approach to improve skin clarity and health. It primarily acts by reducing the activity of tyrosinase, an enzyme crucial for melanin production, and by inhibiting the activation of melanocytes, the cells responsible for pigment synthesis. This dual action significantly curtails the production and transfer of melanin to keratinocytes. Furthermore, it downregulates inflammatory cytokines such as IL-6, IL-8, and TNF-α, thereby suppressing inflammation-induced melanogenesis. The peptide also interferes with UV-induced signaling pathways (p53–POMC–α-MSH–MC1R cascade), which reduces cAMP and subsequently inhibits the PKA–CREB/MITF pathway, further dampening melanin production. Beyond pigmentation control, Tetrapeptide-30 exhibits anti-inflammatory properties, stimulates collagen synthesis, accelerates cellular turnover, improves skin elasticity, and provides antioxidant protection, contributing to a plumped, smoother complexion.

To maintain optimal stability, Tetrapeptide-30 should not be exposed to temperatures exceeding 40°C (104°F). In O/W emulsions, it is best incorporated during the cooling phase below 40°C or directly into the water phase for cold-process formulations. For W/O emulsions, addition prior to homogenization below 30°C (86°F) or into the water phase of cold-process systems is recommended.
