Mechanism of Action
As a precursor to essential ceramide molecules, Phytosphingosine HCL is enzymatically converted upon topical application to fortify the skin's lipid matrix within the stratum corneum. This process significantly strengthens the natural moisture barrier, reducing transepidermal water loss (TEWL) and boosting the skin's natural moisturizing factor (NMF). Beyond barrier support, it exhibits targeted antimicrobial action against *Propionibacterium acnes* and provides anti-inflammatory effects, effectively soothing irritation and minimizing redness.

Key findings
- 01 Phytosphingosine HCL effectively reduces the signs of acne, inhibits the growth of micro-organisms on the skin, and diminishes redness and inflamed skin, demonstrating activity at very low concentrations.
- 02 Phytosphingosine (the base form) at 5 µM significantly increased cornified envelope (CE) production by approximately 1.9-fold and elevated PPARγ mRNA levels by 3.8-fold after 24 hours in normal human epidermal keratinocytes (NHEK cells), suggesting a role in skin differentiation and barrier formation.
- 03 Phytosphingosine (the base form) exhibited an antimicrobial effect against salivary inoculum at a minimum inhibitory concentration (MIC) of 37.5 µg/mL.

Safety
Safety Profile
Phytosphingosine (the base form) has been assessed for safety in dermal cosmetic products at concentrations up to 0.1%. Although a specific CIR report on Phytosphingosine HCL was not found, the CIR Expert Panel generally considers ceramides and their derivatives safe within established cosmetic use practices. Side effects like skin irritation or allergic reactions are uncommon, though patch testing is advisable for individuals with sensitive skin. Data regarding topical use during pregnancy and breastfeeding are currently lacking, and consultation with a healthcare professional is recommended.
