Mechanism of Action
Nonapeptide-1 acts as a competitive antagonist to alpha-melanocyte-stimulating hormone (α-MSH). By binding to the MC1-R receptor on melanocytes, it inhibits the receptor's activation, thereby disrupting the melanin synthesis pathway. This action specifically inhibits tyrosinase, a key enzyme in melanin production, and downregulates the expression of tyrosinase, TRP1, TRP2, and MITF, resulting in an approximate 33% reduction in overall skin pigmentation. Crucially, it maintains essential melanogenesis and exhibits no cytotoxicity. Additionally, early data suggests it may promote collagen synthesis.

Key findings
- 01 A 0.01% concentration demonstrated a 6.42% increase in skin lightening (ΔL) and a 5.39% improvement in overall skin state (ΔE) after 56 days, following 28 days of twice-daily application.
- 02 A 0.0025% concentration enhanced skin luminosity by 3.5% and reduced wrinkles by 15.4% over 28 days in a trial involving 41 women.
- 03 Consistent application over 28 days significantly reverses hyperpigmentation, promoting an equalized skin tone and improved overall appearance.
- 04 Hypothesized to deliver a noticeable skin lightening effect of at least 33%, with potential for continued improvement over time.

Safety
Safety Profile
Nonapeptide-1 is generally well-tolerated for topical application across most skin types, including sensitive skin, presenting a low risk for irritation or adverse reactions. It shows no obvious risks concerning cancer, developmental & reproductive toxicity, or allergies & immunotoxicity. However, data regarding its topical usage during pregnancy and breastfeeding are currently insufficient. While the FDA has not approved labeling for a specific drug product containing Nonapeptide-1, this refers to a drug product, not its use as a cosmetic ingredient. Patch testing is recommended for individuals with known sensitivities.
