METHYL NIACINAMIDE CHLORIDE.

Valuable CAS 1005-24-9 / SKIN CONDITIONING, SKIN PROTECTING

Methyl Niacinamide Chloride (1-MNA) is an active, endogenous metabolite of nicotinamide, distinguished by its anti-inflammatory and vasodilatory properties. It acts to enhance nitric oxide bioavailability and reduce oxidative stress, serving as a beneficial skin conditioning and protecting agent. Clinical findings indicate its potential to alleviate symptoms of chronic dermatoses such as rosacea.

Science

As an active endogenous metabolite of nicotinamide, Methyl Niacinamide Chloride (1-MNA) mediates its effects primarily through increased nitric oxide (NO) secretion and bioavailability, alongside a reduction in endothelial nitric oxide synthase (eNOS)-dependent oxidative stress. This mechanism contributes to its vasodilatory and anti-thrombotic activities. In vitro studies further support its anti-inflammatory efficacy in endothelial cells at concentrations ranging from 0.05-0.2 μM. Beyond these cellular actions, 1-MNA functions directly as a skin conditioning and protecting agent.

Stability

Methyl Niacinamide Chloride is water-soluble. Its optimal stability range is within pH 4 to 6, mirroring its parent compound, Niacinamide. Outside this acidic-to-slightly-acidic range, particularly in alkaline environments, there is an increased risk of conversion to nicotinic acid, which is associated with skin irritation and flushing. Furthermore, exposure to high temperatures exceeding 60°C can compromise its stability.

Safety

CIR Status
Safe as used
Sensitization risk Low

Methyl Niacinamide Chloride is recognized as safe for cosmetic use by the CIR Expert Panel. While no specific maximum topical concentration for Methyl Niacinamide Chloride itself is provided, its parent compound Niacinamide is deemed safe by CIR at concentrations up to 3% in certain product categories. Clinical evaluations of Niacinamide indicated no stinging at concentrations up to 10% and no irritation at concentrations up to 5%. The European Food Safety Authority (EFSA) has also concluded 1-MNA chloride as safe for use as a novel food ingredient up to 58 mg/day for healthy adults, though excluding pregnant and lactating women for ingestion.
