Science

This oil acts via multiple mechanisms to bolster skin health. It delivers powerful anti-inflammatory and antioxidant effects, effectively mitigating oxidative stress and protecting against cellular damage. It significantly improves skin barrier function by reducing trans-epidermal water loss (TEWL) and boosting both keratin water and sebum content, leading to enhanced moisturization. Furthermore, it offers ancillary UV absorption and protection against UVB-induced damage (though not a substitute for sunscreen), and supports anti-aging by promoting skin cell metabolism and potentially inhibiting glycation. For specific concerns, it modulates keratinocyte activity in conditions like psoriasis and suppresses Th2-mediated immune responses, including inhibition of the PI3K-Akt signaling pathway, relevant for atopic dermatitis. The oil also exhibits antimicrobial properties beneficial for acne and promotes effective wound healing.

Key findings

  1. 01 Lithospermum Erythrorhizon Root Extract, at concentrations between 0.002% and 0.02%, was shown to improve skin barrier integrity by reducing trans-epidermal water loss (TEWL), soothe skin, and decrease oxidative stress, also offering potential protection against UVB-induced photoaging.
  2. 02 An emulsion containing 5% Lithospermum Erythrorhizon Root Extract demonstrated a time- and dose-dependent moisturizing effect on human skin, resulting in a maximum increase of 11.77% ± 1.18% in skin humidity.
  3. 03 Traditional Chinese Medicine (TCM) formulations with Lithospermum erythrorhizon as a key herb markedly improved psoriasis symptoms, enhancing skin keratin water content and sebum levels while reducing TEWL, thereby strengthening the skin barrier and immune functions.
  4. 04 Lithospermum erythrorhizon oil significantly alleviated Atopic Dermatitis (AD) symptoms in mice, reducing inflammatory markers (IL-4 and IL-13 levels) and inhibiting the PI3K-Akt pathway by decreasing p-PI3K and p-Akt expression.

Stability

Optimal stability is observed between pH 4.5 and 6.5. Exposure to alkaline systems (pH > 7) should be avoided, as shikonin derivatives present in the oil can rapidly undergo color changes (red to violet/brown) and structural degradation, compromising efficacy. The oil is inherently UV-sensitive, prone to photo-oxidation and polymerization upon light exposure, which can lead to darkening and reduced activity; therefore, UV-blocking packaging and co-formulation with antioxidants are highly recommended. To preserve bioactivity, it should be integrated into emulsions during the cool-down phase, specifically below 40-45°C, to avoid higher processing temperatures. In anhydrous systems, minimize prolonged heating and oxygen exposure. Proper solubilization into suitable lipophilic agents and combination with lipophilic antioxidants are critical to prevent undesired polymerization. While heat tolerant up to 80°C, prolonged exposure at this temperature should be minimized.
